Production of Indole Acetic Acid by 5-fluro-tryptophan Resistant Mutant of Rhizobium Sp. Isolated from Root Nodule of Pulse Legume Phaseolus Mungo L
نویسنده
چکیده
A symbiotic strain was isolated from the root nodules of the legume pulse Phaseolus mungo L and designated as PM 25. The strain was identified as Rhizobium sp. on the basis of the physiobiochemical characteristics and 16S rDNA sequence based homology. The Minimum Inhibitory Concentration of the analogue 5-fluro-tryptophan was 3.0 mM. The analogue (5FT) resistant mutants were screened from wild PM 25 strain after mutagenesis with N-methyl-N ́-nitro-N-nitrosoguanidine (NTG) as a mutagenic agent. The analogue resistant mutants were isolated from 5.0 mM and 10.0 mM 5-fluro-tryptophan supplemented Bergersen’s medium. These mutants produced tryptophan and IAA in Bergersen’s medium without supplemented with tryptophan. The wild type PM 25 was unable to produce tryptophan and IAA in tryptophan nonsupplemented medium but capable of producing IAA in tryptophan supplemented media. So, this Rhizobium sp. is able to follow tryptophan dependent pathway and able to convert tryptophan to IAA.

Twenty six Rhizobium strains were isolated from root nodules of Sesbania sesban (L.) Merr. collected from different regions of Andhra Pradesh. All the 26 Rhizobium strains produced indole acetic acid (IAA), but maximum amount was produced by only five strains in yeast extract mannitol (YEM) medium supplemented with L-tryptophan. The strains were found to elaborate maximum IAA when fed with 2.5...

The Root nodules of Cajanus Cajan plant produced high amount of (35.56 ug/gm) of Indole acetic acid from Hg plants as compared to the N plant. The IAA was measured maximum from Rhizobium spp. of C.cajan plant at 24h, when the bacteria reached the stationary phase of growth. Mannitol & KNO3 preferred to be a good carbon and nitrogen source for IAA production in all N, Ng, H & Hg Rhizobium strain...

A symbiont, isolated from root nodules of Crotalaria saltiana Andr., was identified as Rhizobium sp. It produced large amount of extracellular polysaccharides (EPS) in broth culture in yeast extract mannitol (YEM) medium. The EPS production and growth started simultaneously, though they have different stationary phases. The symbiont produced maximum EPS (16 μg/mL) when the medium was supplement...

Nodulations of Glycine max L. seedlings by parental Rhizobium japonicum (USDA strain 26) and a tryptophan catabolic mutant (tan 4b, NRRL strain B-14075) were evaluated for effectiveness. Seedlings grown in plastic pouches were compared based on increases in root weight (wt.), nodule volume (vol.) and acetylene-reducing (AR) activity. The tan 4b mutant, which can produce extracellular indol-3yl-...
